The participants underwent brain positron emission tomography (PET) scans, body magnetic resonance imaging (MRI), metabolic evaluations, and lipid panels to assess the relationship between obesity, body fat distribution, and Alzheimer’s disease pathology. The researchers measured the volume of subcutaneous and visceral fat using abdominal MRI and examined the impact of these fat deposits on amyloid and tau deposition in the brain.
The results of the study revealed that visceral fat played a significant role in enhancing amyloid formation, explaining a substantial portion of the link between high BMI and Alzheimer’s pathology. The study also found that increased visceral fat was associated with higher levels of amyloid and tau proteins in the brain, characteristic of Alzheimer’s disease. By demonstrating these findings in midlife individuals, the researchers highlighted the importance of early interventions to address obesity-related risk factors for Alzheimer’s disease.
The study also identified associations between brain amyloid deposition and insulin resistance and high-density lipoprotein (HDL) cholesterol levels. Higher levels of HDL cholesterol were found to attenuate the impact of visceral fat on amyloid pathogenesis, suggesting that metabolic and lipid abnormalities play a key role in Alzheimer’s risk in individuals with increased body fat. Moving forward, Dr. Dolatshahi and her colleagues plan to further investigate the relationship between obesity, visceral fat, and Alzheimer’s disease. They will be presenting additional findings at the Radiological Society of North America’s annual meeting in 2024, focusing on how obesity and visceral fat impact cerebral blood flow in midlife individuals. By identifying how visceral obesity affects brain health and function, the researchers aim to provide insights that can inform public health policies and interventions to reduce the burden of Alzheimer’s disease.
